Corporate Overview

Proteostasis Therapeutics, Inc. is the first company dedicated to the discovery of new therapeutics based upon understanding the body’s natural Proteostasis Network.  Advances in our ability to characterize and pharmacologically control the Proteostasis Network create new opportunities for ameliorating diseases in a wide number of therapeutic areas.  Regulation of the Proteostasis Network is a new approach to drug discovery that will provide the kind of foundational understanding that we have witnessed with other breakthrough treatment paradigms, such as with kinases and G-protein coupled receptors.

The Proteostasis Network is an interacting collection of hundreds of proteins controlled by known biological signaling pathways and processes.  Functions of the Proteostasis Network include folding required for protein function, degradation of misfolded or misassembled proteins, and the transport of proteins to appropriate locations in the cell.  If the Proteostasis Network is compromised, every cell type in the body can potentially be affected resulting in eventual disease manifestation.  Using sophisticated tools, the Company has begun to fully map out the Proteostasis Network and use an understanding of its organization to enable the discovery and development of new therapeutics.  We call these drug candidates Proteostasis Regulators, as they are designed to precisely augment or reduce the capacity of the innate Proteostasis Network.

Proteostasis Therapeutics is based on discoveries emerging from the academic laboratories of its scientific founders:  Andrew Dillin of The Salk Institute for Biological Studies, Jeffery Kelly of The Scripps Research Institute and Richard Morimoto of Northwestern University. We have also assembled a broad group of scientific advisors who are thought leaders representing the various components of the Proteostasis Network and associated biological pathways.

In August 2008, we raised $45 million in a Series A round of financing with a syndicate of leading venture capital investors.
